Metabolic Engineering for Robust Natural Product Biosynthesis in C. glutamicum
In this work, a pathway within C. glutamicum was to be engineered to produce a flavonoid naringenin. Towards this, as a first step, 4 different plasmid vectors must be constructed with the genes CHSph and 4CLpc, from P. deltoides; CHSat and 4CL2at, from A. thaliana; and CHI, from M. sativa. The process employs a Polymerase Chain Reaction (PCR) to amplify the genes of interest, followed by extraction of the desired segments. Gibson Assembly was performed to assemble the fragments and with this approach we were able to construct 2 of the plasmid vectors.
