Enhancing Self-Contemplation Techniques and In-context Training for T Cell Receptor Generation for Novel Epitopes Using Language Models

This research aims to generate and evaluate computationally designed T cell receptors (TCRs) for novel epitope binding to advance the development of personalized immunotherapy. A pre-trained protein language model, RITA, is fine-tuned using in-context training and self-contemplation prompting (SCP) to generate high-quality TCR sequences. Evaluation is conducted using various tests to assess binding affinity and similarity to natural TCRs, ensuring the reliability of generated sequences. By reducing reliance on costly wet lab validation, this approach accelerates T cell engineering and improves the efficiency of therapeutic design for immunotherapy applications.
